Introduction Colorectal cancer (CRC) remains a significant health challenge, with increasing incidence and mortality rates worldwide. The advent of Compound Kushen Injection (CKI), a traditional Chinese medicine, offers a promising adjunct to conventional chemotherapy. This blog delves into the findings of a systematic review and meta-analysis that evaluates the efficacy and safety of CKI combined with chemotherapy in treating advanced CRC.
Study Overview The systematic review and meta-analysis conducted by Wu et al. encompassed 16 randomized clinical trials, involving 1378 patients with advanced CRC. The primary aim was to ascertain the clinical benefits and safety of combining CKI with chemotherapy compared to chemotherapy alone.
Efficacy of CKI The meta-analysis revealed significant improvements in several key clinical outcomes when CKI was used alongside chemotherapy:
- Objective Response Rate (ORR): The combination therapy increased the ORR by 30% (RR = 1.30, 95% CI: 1.18-1.44).
- Disease Control Rate (DCR): A modest but significant improvement in DCR by 8% (RR = 1.08, 95% CI: 1.03-1.13) was noted.
- Performance Status: Patients receiving CKI exhibited a better Karnofsky Performance Status (KPS) improvement rate (RR = 1.18, 95% CI: 1.07-1.31).
Safety Profile CKI also demonstrated a favorable safety profile by reducing the incidence of several chemotherapy-induced adverse reactions:
- Leukopenia: Reduced by 26% (RR = 0.74, 95% CI: 0.62-0.87).
- Thrombocytopenia: Reduced by 32% (RR = 0.68, 95% CI: 0.49-0.94).
- Gastrointestinal Reactions: Reduced by 28% (RR = 0.72, 95% CI: 0.55-0.94).
- Liver Damage: Reduced by 52% (RR = 0.48, 95% CI: 0.30-0.79).
Immune Function The analysis also indicated improvements in immune function markers:
- CD4+ T Cell Ratio: Increased significantly (MD = 9.70, 95% CI: 8.73-10.68).
- CD4+/CD8+ Ratio: Improved (MD = 0.25, 95% CI: 0.22-0.28).
- CD8+ T Cell Ratio: Decreased (MD = -5.25, 95% CI: -5.94 to -4.56).
Conclusion The findings underscore the potential of CKI as a valuable adjunct to chemotherapy in treating advanced CRC. CKI not only enhances therapeutic efficacy but also mitigates the adverse effects associated with chemotherapy, thereby improving overall patient outcomes. However, further large-scale, multi-center RCTs are essential to corroborate these findings and establish standardized treatment protocols.
